How Micellisation Works

WHY THE FORMAT MATTERS

Delivery systems are often described as if they were interchangeable, and they are not. A micelle, a liposome and a vesicle are built differently and sit at very different sizes, and that determines what each one can carry and how the body handles it. Protocol builds micelles: the smallest of the three, with a single shell around an oil-soluble core. Here is how they compare.

Micelle 5–300 NM

Single shell. An oil-soluble core inside one water-soluble sheath. Diameter varies with how much active is loaded. Protocol builds these from gum arabic, a plant-derived raw material.

Liposome 50–2,000 NM

Double membrane, hollow. A bubble enclosed by two layers of phospholipid, with a watery interior rather than an oil core, a different architecture for a different payload.

Vesicle 800–1,500 NM

Cellular machinery. A double-membrane compartment cells use to move cargo in and out and to hold reactions apart. It is biology's own transport system, not a formulation format.

Malaria remains one of the world's heaviest and most preventable disease burdens, and it falls hardest on the places least able to store, ship, and administer conventional medicine. Protocol plans to begin its humanitarian work in the central and eastern region of the continent, then expand outward from there, with formulations engineered for the realities of field distribution.

WHY IT PERSISTS

Malaria is treatable. It continues at this scale because treatment has to survive the journey to the person who needs it. Much of the burden falls in rural areas that are hours from a clinic, where refrigeration is unreliable and supply chains are long, and where a course of treatment is often started but not finished.

Protocol is building for those conditions rather than around them. A formulation that holds for three years without a cold chain, dosed as oral drops for infants through adults, at a lower dose for the same effect. The aim is a treatment that reaches the last mile intact and is simple enough to be taken to completion.
